GENERAL SUMMARY/OVERVIEW STATEMENT:

Under supervision, and within established departmental and hospital policies and procedures, is responsible for performing a variety of general and technical duties to support the research activities within the Center for Neurologic Diseases.

PRINCIPAL DUTIES AND RESPONSIBILITIES:

Principal Duties:

  • To obtain genotyping information for rodents and aid in animal husbandry.

  • To assistant in culturing and analyses of human stem cells.

Research Activities:

  • Performs tagging and tailing of mice, DNA extractions, PCR, and organizes colonies.

  • Performs tissue culture duties and uses molecular biological techniques to analyze cellular phenotypes.

  • Performs literature searches as applicable to research projects.

Laboratory Activities:

  • Performs tagging and tailing of mice, DNA extractions, PCR, and organizes colonies.

QUALIFICATIONS:

  • Currently enrolled in a Bachelors program.

  • Course work in physical and biological sciences.

SKILLS/ABILITIES/COMPETENCIES REQUIRED:

  • Must have demonstrated ability to learn quickly and understand scientific concepts, and the ability to work in a team environment.

  • Computer Literacy.

  • Knowledge of research protocols ability to methodically record data and analyze data using statistics.

  • Good interpersonal and communication skills.

  • Ability to perform routine tests and to prioritize work.

WORKING CONDITIONS:

  • Laboratory environment.

  • May be exposed, under controlled conditions, to radioisotopes and other hazardous materials.

FISCAL RESPONSIBILITY:

Will be responsible to maintain supplies as necessary to complete assigned duties.

VIEW JOBS 11/14/2019 12:00:00 AM 2020-02-12T00:00 The Technical Research Assistant II will be working in the Gene-editing in Regenerative Medicine Laboratory at the Brigham and Women's Hospital under the direction of the PI, Dr. Shailesh Agarwal. The mission of the laboratory is to demonstrate a pathway to translate gene-editing from in vitro experiments to small- and large-animal studies. We are seeking a qualified research associate with significant in vitro experience with gene-delivery and gene-editing. He/She must be able to work independently, organize research experiments, and be able to function as a lab manager in addition to scientific tasks. PRINCIPAL DUTIES AND RESPONSIBILITIES: * This individual should be experienced in the following approaches: * Cell culture * Tissue-derived cell culture and cell lines * Maintenance * Proliferation and apoptosis assays * Differentiation assays * Gene delivery (electroporation and adenoviral, possible nanoparticle) * Into Cells in vitro/ex vivo * Into Tissues ex vivo * Plasmid construction and modification * Swapping transgene cassettes in plasmids * In vitro/ex vivo CRISPR-based approaches * Gene-cleavage * CRISPR interference * Homology-directed recombination * Gene-editing assays * Cleave efficiency * Off-target assays * Immunoblot assays (Western blot, ELISA) * Flow cytometry (sample preparation, staining, and machine use) * Perform a high volume of experiments related to the above techniques efficiently, effectively, and without requiring daily direction from the PI. * Maintain clear and detailed laboratory records, as directed (paper and electronic). * Provide professionally documented figures to be used for grants, presentations, and manuscripts. Maintain professional and timely communication with all academic and industry collaborators and other stakeholders. SUPERVISORY RESPONSIBILITIES: Responsible for training and orienting new staff, as needed. QUALIFICATIONS: BS in biological/physical science required. At least two (2) years of experience in a directly related lab research setting required. Experience with in vivo In vivo gene delivery (e.g. tissue electroporation, nanoparticle, or AAV delivery) Murine bone marrow transplantation SKILLS/ABILITIES/COMPETENCIES REQUIRED: High degree of computer literacy. * Sound analytical and organizational skills. * Demonstrated ability to analyze data statistically and to logically present it. * Good oral and written communication skills. * Must be able to logically and effectively structure tasks and set priorities. * Ability to identify potential problems and trouble-shoot solutions. * Ability to maintain an open mind, energy, and enthusiasm for the work. * A 1-year commitment is requested; a 2-year commitment is appreciated.
