Ionis Pharmaceuticals, Inc., located in the North San Diego County city of Carlsbad, was founded in 1989 and was purposely designed to create a better, more efficient drug discovery platform, establish a new innovation-centered business model, and maximize the value of every medicine created and get it to the people who need it most as quickly and as efficiently as possible.

With the RNA molecule as the basis of our novel drug discovery platform, we are targeting an unprecedented range of therapeutic areas. Our antisense therapies are disrupting diseases and changing their coursefrom the rarest of conditions, to those that impact millions of people. We currently have three commercial products and more than 40 potentially transformative medicines in our pipeline designed to treat a broad range of diseases.

Ionis is a challenging, motivating and rewarding environment designed to foster innovation and scientific excellence. Our success is a direct result of our outstanding employees. We are interested in bringing together individuals with different skill sets into this creative environment to be part of a productive team. Join us and experience our unique culture while you develop and expand your career.

The selected candidate will work with a multi-disciplinary team of energetic researchers who are focused on unraveling the complex mechanisms underlying antisense oligonucleotide (ASO) drugs. The emphasis will be to understand mechanisms of ASO action in cells and intracellular trafficking pathways of ASOs.

  • Responsible for investigating how ASO drugs work in cells, with a goal to improve the drug design and to expend antisense technology

  • Specifically studies how ASO drugs enter and traffic in cells, the ASO release mechanisms from endosomes, intracellular action sites of ASOs, interactions of ASOs with cellular proteins and RNAs, and the consequences of such interactions on the performance of the ASO drugs and biological processes

  • Identify ways to improve efficient ASO release from endosomes

  • Emphasis on understanding the mechanisms of ASO-directed, RNase H cleavage of target RNAs

  • Develop new antisense mechanisms to modulate gene expression with clinical application

  • Works with a multi-disciplinary team of energetic researchers who are focused on unraveling the complex mechanisms underlying antisense drugs

  • Conducts studies to understand the mechanisms of how antisense drugs work in cells using different techniques including but not limited to cell culture and small animal handling, confocal imaging, DNA/Protein and essentially RNA manipulation, gene expression manipulation, structure-activity relationship study, protein/RNA/Oligonucleotide subcellular localization and trafficking analysis using both biochemistry and cell biology approaches

  • Present and publish research data

  • A Ph.D. in Cell Biology, Molecular Biology, Biochemistry, or related field

  • At least 3 years of post-doctoral research experience, and previous industry working experience (>2 years) is a plus

  • Broad knowledge and experimental experience with nucleic acid-protein interactions, RNA biology, vesicular trafficking, membrane and cytoskeleton are desired

  • Track record of innovative research and scientific publications in the related field

  • Skills/experience in small animal handling, confocal imaging, cell biology, and antisense technology are highly preferred

  • Excellent organizational, interpersonal and oral communication skills

  • An ability to work both independently and as a team member, and be productive and successful in an intense work environment

Excellent salary and benefits package offered.

VIEW JOBS 4/19/2019 12:00:00 AM 2019-07-18T00:00 Job ID : 86921BR Location : US - California - Carlsbad : Job Description When you join us at Thermo Fisher Scientific, you'll be part of a smart, driven team that shares your passion for exploration and discovery. With revenues of more than $22 billion and the largest investment in R&D in the industry, we give our people the resources and opportunities to make significant contributions to the world. How will you make an impact? This scientist will be working at the forefront of cell biology, developing integrated applications using a wide array of Thermo Fisher Scientific products, particularly stem cells and genome engineering. This work will support three principal activities, to exemplify how to use our tools to ask broad scientific questions, to execute custom services for our customers and to participate in external collaborations. The scientist will be expected to lead projects independently and to work with junior scientists in a highly collaborative team environment. Projects may include, but not be limited to, cell line engineering, high-throughput or high-content assay development, reprogramming of adult somatic cells to induced pluripotent stem cells (iPSCs), genomic editing of iPSCs, differentiation of iPSCs and the use of CRISPR libraries to execute functional genomics screens. These projects may be driven by internal project design or by external collaborations. What will you do? The successful individual will directly interact with senior R&D leadership, project management and external collaborators to develop workplans. The scientist will then be responsible for executing these plans to meet agreed upon deliverables and timelines. These activities will require a significant level of independence but may also be combined with activities of either other scientists on the team. This role will involve the following tasks: * Molecular biology: Plasmid subcloning, RT-PCR, TaqMan® PCR, western blotting, sequencing * Stem cell culture (feeding, passaging and cryopreservation) * Genomic editing (TALN and CRISPR based editing) * Stable cell line generation (transfection and antibiotic-selection) * Assay development (high-throughput and high-content) * CRISPR Screens (array-based CRISPR screens in cell-based assays) * Periodic weekend work (stem cell culture requires daily attention) * Because stem cell culture requires the daily feeding and observation of cells, weekend work will occasionally be required depending on the nature of active projects. * This individual will also be expected to support and participate in senior level meetings, business reviews, etc. How will you get here? Education/Experience * Requires a PhD degree in the life sciences. Candidates with a bachelor's degree (10+ years experience) or a master's degree (7+ years experience) may be considered. * Requires 1-5 years experience with: o Stable cell line generation o Stem cell culture o Genome engineering * Prefer 1-5 years of experience with: o Assay development o Molecular biology techniques (DNA/RNA/protein) Knowledge, Skills, Abilities * Perform standard laboratory math calculations and solutions/media preparation * Manipulating DNA plasmid constructs, PCR, western blots * Sterile tissue culture technique, particularly stem cell culture in the absence of antibiotics * Assay development (particularly fluorescent based assays) * Genome engineering * Use of CRISPRs for genome editing and/or screening * Formal training in stem cell culture * Observe deviations from the expected outcomes * Succeed with a moderate level of supervision and guidance * Work within a strong team with demonstrable influence/interpersonal skills At Thermo Fisher Scientific, each one of our 70,000 extraordinary minds has a unique story to tell. Join us and contribute to our singular mission—enabling our customers to make the world healthier, cleaner and safer. Apply today!
