Position Summary:

We are seeking an independent and self-motivated Senior/Principal Scientist to be a project leader in our CAR T cell construct engineering group. The main goals for this position is to help establish our construct engineering pipeline, integrating streamlined construct generation, testing and validation. This candidate will also help develop ways to utilize gene editing into our pipeline for site directed construct integration. The ideal candidate will integrate new ideas to this pipeline for high throughput cloning, viral and non-viral library generation and automated screening. We would like candidates with previous experience in viral construct engineering, a strong molecular biology background and initiative to dive into a fast-paced new startup and be creative.

Requirements

Essential Functions:

  • Lead the development of our construct engineering and screening platform for CAR T cell constructs for viral and non-viral delivery
  • Develop ways to automate our cloning and plasmid production for high throughput evaluation of a large range of constructs
  • Manage junior scientists in the lab and aid in their experimental design and execution
  • Willing to wear many hats as is expected in a start up environment
  • Work in a collaborative, multidisciplinary team and help mentor others

Preferred Experience:

  • Extensive experience with both basic and advanced recombinant DNA cloning techniques, including restriction cloning, Golden Gate assembly, Gibson assembly, and PCR-based assembly
  • Demonstrated hands-on experience with molecular techniques, including oligonucleotide and gene fragment design and synthesis, quantification and isolation of DNA and RNA, PCR genotyping, sequencing analysis, media preparation, bacterial culture, sterile technique
  • Extensive knowledge of virus titration methods, copy number determination, integration site analysis and molecular biology techniques such as qPCR and flow cytometry is required
  • Knowledge of genome editing technologies, gRNA design and verification
  • Experience with multi-color flow cytometry and cell sorting for construct testing
  • Familiarity with Geneious, Benchling, or other software for plasmid design and sequence analysis
  • Experience with integrating automation into molecular biology pipeline
  • Great attention to detail, resourceful, and well organized
  • Previous experience managing junior staff
  • Outstanding written, visual, and oral communication skills with a willingness to have ideas challenged and to challenge others ideas in a constructive, efficient and respectful way
  • Start-up /industry experience a plus

Preferred Education:

  • PhD and 8+ years (or equivalent) experience in molecular biology, bioengineering or related discipline with relevant lab comprehension

Preferred Additional

Skills:

  • Previous experience utilizing viral vectors for gene delivery (preferably in the setting of T cell biology)
  • Preparation and maintenance for process development and research grade plasmid production operations, including SOPs, batch records, reports, and supporting regulatory filings
  • Prior experience working with human samples, specifically primary T cell culture
  • Experience with Electronic Lab Notebook (ELN) systems
  • Demonstrated project management skills
