What You Will Achieve We are seeking a Research Scientist with a breadth of molecular and cell biology skills including assay development experience to join the Cellular, Genomic, and Protein Sciences (CGPS) group within Pfizer. The CGPS group is responsible for a range of functions including target identification and validation, establishment of cellular and protein reagents, and development and use of screening assays to help advance early discovery programs. The focus of this role will be to evolve our mRNA vaccine platform capabilities, while simultaneously generating data to advance ongoing vaccine programs. How You Will Achieve The successful candidate will join a team of scientists within the Cellular, Genomic, and Protein Sciences group (CGPS) that is responsible for a range of functions including target identification and validation, establishment of cellular models and protein reagents, and development of assays to help advance early discovery programs. This individual will apply strong expertise in molecular biology methods and assay development skills to advance the mRNA vaccine portfolio. Focus will be on addressing overarching challenges associated with mRNA therapeutics, as well as program-specific efforts to select and advance existing vaccine candidates. Successful Candidates will be expected to: * Develop and implement a suite of comprehensive assays to evaluate and optimize mRNA vaccine candidates. * Apply cloning, expression plasmid design, in vitro transcription, transfection, and cell-based assays to mRNA optimization efforts. * Help to identify, evaluate, and, where applicable, implement novel methods or technologies that would enhance workflow efficiencies and/or improve the quality of our mRNA vaccine platform. * Analyze and interpret data, prepare cogent data summaries, and communicate results to partners in the Research Units and Partner Lines. * Maintain up-to-date electronic laboratory notebooks and records in accordance with Pfizer policy, and ensure proper archiving of banked cell vials. * Be highly motivated, organized and show ability to work effectively in a team-oriented environment. Qualifications Must-Have * BS with 3+ of relevant laboratory experience OR MSc with 0+ experience. * Expertise with standard methods for RNA/DNA isolation, quantification, and QC analysis. * Hands-on experience with various molecular biology techniques including DNA cloning and expression vector design, in vitro transcription for mRNA synthesis, quantitative PCR, qRT-PCR and ddPCR, primer/probe design, and methods for modulating gene expression (RNAi/CRISPR, or overexpression). * Hands-on experience with mammalian cell culture and development of cell-based assays, and methods for reagent delivery (ie: electroporation or lipofectamine transfections, lentiviral delivery). * Proficiency with standard methods for gene/protein expression analysis including mRNA (qRT-PCR) and protein analysis, Western blotting, immunostaining, and FACs. * Experience designing and conducting in vitro pharmacology assays to study cellular endpoints in response to small molecule compounds and/or other agents. Nice-to-Have * Development of engineered, clonal cell lines * Experience with any of the following cellular assay formats: ddPCR, gene reporter assays, cytokine secretion, HTRF and/or alphaLISA. * Familiarity with basic next-generation sequencing methods. * Previous hands-on experience with plate-based robotic automation for RNA/DNA isolation and QC, and/or library preparation. * Prior experience and knowledge of mRNA vaccine development. * A background in the field of Virology. PHYSICAL/MENTAL REQUIREMENTS * Sitting, standing, lifting, bending, and walking. * Ability to perform practical laboratory bench work and work on a computer (while standing or sitting). * Ability to perform mathematical calculations and interpret data.
