This team is driving innovative transcriptome gene editing of disease causing RNA with tunable control for safe and effective treatments for patients with rare diseases. In this role you will work from the bench with in vitro molecular and cellular techniques to develop target engagement assays to systematically test novel applications of a novel gene editing platform. You will also have a seat at the table and work to devise platform focused research strategies and process improvements. In This Role You Will:

  • Work in the lab to develop assays to facilitate testing experimental hypothesis on novel platform

  • Testing studies using cultured cells employing a variety of cellular and molecular assay technologies

  • Independently execute studies, analyze data, share the conclusions and propose follow up experiments with team and founders

  • Design and clone novel DNA constructs using state-of-the-art cloning strategies and techniques

  • Explore viral or non-viral delivery strategies for intracellular cargo and payload strategies

  • Examine data and prepare technical reports, summaries, and present at team meetings

You Will Be Required to Bring:

  • Ph.D. in Life sciences with 2+ years of industry or Postdoctoral experience with a strong publication record in CRISPR related technologies or gene editing

  • Significant experience of 3+ years in hands-on molecular cloning techniques

  • Extensive knowledge and experience with molecular cloning and DNA plasmid design, nucleic acid isolation, PCR, qPCR and molecular biology

  • Preference for Scientists who are highly skilled in gene editing

  • Ample experience with mammalian tissue culture and cell-based assay execution

  • Familiarity with DNA/plasmid editing software and genome browsers

  • A strong desire to use technology and creative solutions to address clinical unmet medical need in patient outcomes is necessary

  • A "Biotech mindset" and flexibility in implementing new strategies, cutting-edge technologies, and methods when faced with obstacles in executing platform technologies/studies

  • Ability to communicate best path forward strategies effectively with cross-functional teams, key stakeholders, partners and successfully contribute to platform advancement

Role requires experience in using CRISPR/CAS9 as a tool to develop novel cellular cancer models. The candidate will be a highly motivated experimentalist with a strong desire to succeed at the highest level.</p> <p><strong>Principal Duties and Responsibilities</strong></p> <p></p> <p>Engineer cell lines with knock-out or knock-in of genes of interest using non-intergrated sgRNA and CAS9</p> <p></p> <p>Design strategies for cloning fusion reporter gene vectors, identify CROs to perform cloning and manage execution of the work.</p> <p>Utilizes lasted Molecular biology tools to NGS,ddPCR rtPCR, advance cloning techniques Golden Gate, Gibson Assembly to build genomic knock up down models.</p><p><strong>Requirements</strong></p><p>BS/MS equivalent in cell/molecular biology, cancer biology or genetics with at least 1 year hands-on experience in cancer research preferably in a pharmaceutical setting</p> <p>Experience with molecular biology methods and assays such as molecular cloning, RNA and protein expression approaches (e.g. QPCR, Western blotting) as well as various genetic perturbation methods.</p> <p>Experience in gene editing using CRISPR/CAS9, particularly strategies that use non-interfated sgRNA and CAS9.</p> <p>Ability to maintain timely and detailed notes using electronic lab notebook as well as demonstrated experience using Microsoft Excel and GraphPad Prism </p> <p>Excellent interpersonal skills and willingness to learn a variety of new techniques and technologies; Ability to adapt to rapidly changing priorities and deadlines</p> Monte Rosa Therapeutics Boston MA
