Fulcrum Therapeutics is a clinical-stage biopharmaceutical company focused on improving the lives of patients with genetically defined rare diseases in areas of high unmet medical need. Fulcrum's two lead programs in clinical development are losmapimod, a small molecule for the treatment of facioscapulohumeral muscular dystrophy (FSHD) and pociredir, a small molecule designed to increase expression of fetal hemoglobin for the treatment of sickle cell disease and other hemoglobinopathies. The company's proprietary product engine identifies drug targets that can modulate gene expression to treat the known root cause of gene mis-expression.
Description
Fulcrum Therapeutics is seeking a Research Associate/ Senior Research Associate, who will be a driven professional, eager to bolster our pharmacology research initiatives. This role involves spearheading the design and implementation of in vitro, in vivo, and ex-vivo pharmacology studies, critically contributing to our understanding of in vitro/in vivo correlation, pharmacokinetics/pharmacodynamics (PK/PD), target engagement, and therapeutic efficacy of our novel drug candidates. As a crucial member of our multidisciplinary team, you will have the opportunity to lead and execute pre-clinical discovery studies, generating decisional data packages to identify and support the advancement of clinical candidates.
Responsibilities
Be a key contributor to biology efforts for multiple -centric project teams.
Design, develop, and execute experiments across a diversity of human primary and iPSC-derived cell culture models.
Carry out rodent in vivo studies adhering to IACUC standards, independently and within a team dynamic.
Design and execute studies ranging from early exploration to IND submission stages, focusing on in vivo/ex vivo sample generation and analysis, PK/PD modeling, and therapeutic efficacy.
Performing assays to assess target engagement and biomarker analysis in samples such as blood, bone marrow, and muscle tissue, contributing to our understanding of disease mechanisms.
Develop, optimize and conduct in vitro/ ex vivo assays and molecular biology techniques (RT-qPCR, ELISA, protein assay, and flow cytometry).
Demonstrate independent thought/creativity in science with excellent collaboration and communication across functional disciplines and multitasking skills.
Maintain accurate, timely and complete electronic laboratory notebook entries.
Qualifications
BS or MS with relevant experience in areas related to Cell Biology, Molecular Biology and/or Pharmacology in an academic or industry setting.
Experience working with rodents a must, administering doses through various methods including but not limited to PO, IP, IV, SC and IM, and conducting post-study processes including blood draws, necropsies, and ex vivo sample analysis.
Hands-on experience with in vitro (cell culture) models of diseases.
Experience with Contract Research Organizations (CROs) is an asset.
Hands-on experience with a range of in vitro/ ex vivo techniques (such as RNA/DNA extraction, RT-qPCR, ELISA, protein analysis via Western blot, WES, and/or MSD) is desirable.
Experience running flow cytometry and/or ICC, assays are a plus.
Proven ability to independently plan and execute experiments, analyze data, and present findings.
A passion for contributing to a team-oriented, diverse, and inclusive work environment.
Excellent multitasking abilities and able to thrive in a fast-paced, collaborative setting.
