JOB SUMMARY:

Sangamo Therapeutics is seeking a talented Research Associate in our Discovery and Translational Research group to support the development of our novel ZFP-TF platform for the treatment of CNS diseases.

The successful candidate will work within Discovery and Translational Research and contribute to the development of novel CNS therapeutics. Key responsibilities will include performance of neuronal cell culture experiments to demonstrate target engagement and support lead candidate selection, screening and assay development, and generation of data from in vivo studies to support advancement of CNS programs. The position requires adaptability, creativity, good written and verbal communication skills, as well as a capacity for constant innovation in pursuit of company goals. ESSENTIAL FUNCTIONS:

  • Perform in vitro experiments in primary and iPSC-derived neurons

  • Perform chronic AAV expression studies in mammalian cell models

  • Expertise in molecular and biochemical methods (qRT-PCR, ELISA)

  • Process animal tissue for RNA, DNA, and protein endpoints

  • Successfully plan, execute, and document laboratory procedures and experiments with great attention to detail

EDUCATION, EXPERIENCE AND SKILLS REQUIREMENTS:

  • B.S. or M.S. in molecular and/or cellular biology, biochemistry, neuroscience, bioengineering, chemical engineering, or related discipline

  • 4+ years of relevant laboratory experience in an academic research laboratory or biotechnology company. Level will be determined by education, experience, and skills

  • Experience with neuronal cell culture, including AAV and nucleic acid transfection

  • Extensive experience with molecular biology and biochemical methods and techniques including qRT-PCR, ELISA, western blotting, and nucleic acid and protein isolation from mammalian cells and animal tissues

  • Preferred candidates will be proficient in use of the Microsoft Office Suite and statistical analysis and graphing software (e.g. GraphPad Prism, SigmaPlot)

OTHER QUALIFICATIONS:

  • Strong experiment design, problem-solving, and data analysis skills are desired

  • Excellent communication and presentation skills

  • Experience with high-throughput assays preferred

  • Experience with gene therapy and CNS-related fields preferred

JOB SUMMARY: Sangamo Therapeutics is seeking a mid- to senior-level Research Associate to join our Cell Therapy group, focusing on the development of the next generation of cellular therapies. The successful candidate will contribute to the advancement of our CAR Treg platform. The environment is teamwork-oriented, exciting, fast-paced, and offers the opportunity to directly impact the clinical translation of genome editing. ESSENTIAL FUNCTIONS: * Isolation of primary human T (and ideally Treg) cells and their maintenance in tissue culture. Perform in vitro electroporation, AAV transduction or lentiviral transduction to achieve gene editing and CAR expression. * Isolate genomic DNA, perform PCR amplification and library preparation from genomic DNA extracts for next-generation sequencing (NGS) to determine levels of genome modification and targeted integration. * Perform gene expression analyses by RT-PCR or related methods. * Clone new constructs and produce lentiviral vectors, including production of lentiviruses and titration. * Perform flow cytometry analysis of ZFN-edited cells and/or CAR-expressing cells. Develop robust new panels for phenotyping and other flow-based immunoassays. * Perform functional analysis of edited Treg, including suppression assays * Support lab management and infrastructure. * Document methods, maintain laboratory notebooks and databases. EDUCATION, EXPERIENCE AND SKILLS REQUIREMENTS: * B.Sc. or M.Sc. in a scientific discipline (preferably immunology or molecular and cell biology) with at least 5 years of experience, preferably in an industry setting. Level will be dependent upon education, skills and experience. * Highly proficient with aseptic cell culture techniques and cell-based assays using primary human cells. Experience in Treg cell isolation and functional assessment a plus. * Highly proficient in flow cytometry and experienced in applying rigorous, standardized methods to generate high quality data using FlowJo. * Proficient in molecular methods such as cloning, nucleic acid extraction, PCR. Illumina sequencing experience is a plus. * Proficient in the use of computer programs for sequencing and DNA construct generation, data analysis and assembly (e.g. Geneious or similar, MS Excel, MS Powerpoint).
