What is this role really like?

The Drug Discovery team at Mirati is responsible for the invention, optimization and characterization of quality small molecules as clinical candidates for Mirati research programs. The Principal Scientist will be responsible for medicinal chemistry strategy, compound design, synthesis and compound progression activities enabling advancement of oncology drug discovery programs. Key efforts will be directed at the invention, preparation, and characterization of compounds as potential new targeted anticancer drug candidates; the advancement of compounds through hit-to-lead, lead optimization and candidate selection activities, and initiation of new projects by identifying viable starting points for chemistry activities. Experience with structure-based drug design and optimizing lead compounds within a drug discovery program is desired.

Day-to-Day Duties:

  • Lead a drug discovery project from early stage through candidate selection.

  • Drive medicinal chemistry and lead optimization strategies as a key member of drug discovery teams.

  • Optimize compound delivery and synthetic efficiency, primarily utilizing FTE and fee-for-service based CROs. Maintain effective communication with CROs (email; teleconferences; web meetings), visiting key CROs as necessary. Contribute to building a productive, internal Mirati chemistry lab.

  • Contribute novel IP as a key member of drug discovery teams.

  • Coordinate compound progression activities with outside partners and CROs

  • Maintain corporate database; prepare and deliver internal presentations, patent applications and external publications as driven by the project and corporate strategies.


  • Ph.D with 3+ years of experience in a relevant chemistry discipline in a laboratory environment with synthetic organic chemistry and drug discovery background

  • Relevant experience in a synthetic/medicinal chemistry laboratory, with pharmaceutical or biotech industry experience preferred

  • A proven track record of experience and expertise in synthetic chemistry, medicinal chemistry and drug discovery. Demonstrated experience in advancing lead compounds through preclinical discovery stages to candidate selection preferred

  • Strong understanding of key components and processes of drug discovery

  • Experience with Fragment-based drug discovery, Structure-based drug discovery, fundamentals of cheminformatics, and Property-based drug design preferred

  • Demonstration of outstanding communication skills through internal and external presentations and publications

We develop and co-validate bioanalytical assays to characterize products in biological samples. We are seeking a highly motivated individual with experience in bioanalytical method development to characterize biotherapeutic portfolios from discovery to clinical studies. You will have an opportunity to focus on the discovery and development of state of the art biotherapeutic platforms, such as CAR-T, multi-specifics, focusing on multiple disease areas with the intent of delivering novel therapeutics for patients with few therapeutic options, using partnership across multi-functional teams. The Principal Scientist will join our Biologics Discovery organization located in San Diego. Biologics Discovery identifies and develops lead biotherapeutic molecules for a range of indications and therapeutic areas within Janssen R&D. The Principal Scientist will be leading protein engineering efforts for development of non-antibody protein therapeutics as an independent contributor, closely collaborating with and utilizing support from co-located functional groups. You will propose ideas and define strategies for engineering initial candidate molecules into optimized lead protein therapeutics and drive, coordinate and contribute to the lab work to execute the strategies. You have subject matter expertise in one or more areas that are germane to protein engineering and can deliver results within aggressive but achievable timelines. This person will effectively summarize and communicate project strategy, work, results and conclusions. In a highly matrixed organization, your ability to build productive interactions with colleagues at all levels and from varied functions will lead to being both a strong team leader and team member. You will be a member of multiple therapeutic and new platform/technology development project teams and provide expertise and support needed to help deliver on project goals. You will provide and test ideas for new technologies that can produce novel biotherapeutics or improve their performance and potentially build intellectual property for the organization. Qualifications * Ph.D. degree in Protein Biochemistry, Protein Engineering or related field with 5+ years of experience, preferably in research within the biotechnology or pharmaceutical industries required. * Demonstrated experience with protein design to improve/modify protein function, biophysical properties, production levels or other attributes and with implementation and testing of design is required. * Knowledge and hands-on experience with current innovative or high throughput methods for molecular biology, protein variant libraries, prokaryotic or eukaryotic expression and purification, screening or other that facilitate efficient protein engineering is required. * Experience and aptitude with biologics databases is preferred. * Project leadership experience is a plus. * Experience with structure-based protein design and requisite software is a plus.
