Co-op Research Associate - Biology

Monte Rosa Therapeutics is seeking a passionate Co-op Research Associate to assist in a variety of different technical disciplines. Monte Rosa seeks to eliminate disease causing proteins through targeted protein degradationa novel and exciting way of drugging proteins that until recently were considered un-druggable. The candidate will help on a variety of programs and technical projects from developing and characterize cellular models, or that will be used for target-identification and -validation as well as Assist in execution of a variety of assays to mechanistically understand the target of interest and to screen and characterize small molecule compounds and determine the mode of compound action, or for high-throughput compound screens to identify novel degraders. The ideal candidate is someone looking to learn a variety of cellular, biochemical or molecular techniques, is capable of multi-tasking and working in a team environment, and is curious and enjoys doing science.

Principal Duties and Responsibilities:

Assist in the establishment and characterization of cell lines with specific genetic alterations (gene mutation, gene deletion, gene modifications) using CRISPR/Cas9

Simultaneously and aseptically maintain multiple unique cell lines in culture

Confirm the introduction of the genetic alternations using molecular biology assays (Western Blot, PCR).

Evaluate the effect of the genetic alternation on the cells using Next Generation Sequencing

Help to develop and run a variety of assays to screen and characterize target proteins and small molecule compounds.

Perform a variety of high-throughput Cellular Biology and Biochemical experiments and analytical assays such as ELISA, HTRF, high content, flow cytometry, and reporter assays (fluorescent and enzymatic).

Requirements

  • Currently in a Master degree or equivalent program in Biochemistry, Cell Biology or a life science.

Strong desire to learn the most advanced approaches to drug discovery

Experience with mammalian cell culture

Understanding of molecular biology, biochemistry techniques (e.g. Western Blot, PCR, ELISA).

Enjoys working in a team-based environment

Excellent oral and written communication skills

Combining deep knowledge, insight and leading-edge science, our integrated engine is built for that purpose.</p> <p>Monte Rosa Therapeutics is seeking an energetic and self-motivated scientist to join our innovative team working towards discovery of unique molecular glue degraders for the treatment of targeted patient populations in indications including auto-inflammatory disease, hematology and oncology. This is a lab-based position that offers the opportunity to impact projects as they progress from target validation and hit-finding, through to lead optimization and pre-IND studies.</p> <p>The candidate will be a highly motivated experimentalist with demonstrated collaborative skills and a strong desire to contribute to project success.</p> <p><strong>Principal Duties and Responsibilities</strong></p> <ul> <li>Validate molecular glue degrader hits by employing techniques such as Western blot, flow cytometry, ELISA, qPCR to assess viability, target and biomarker expression, proliferation.</li> <li>Develop, validate and independently execute cell-based assays using cell lines and primary human hematopoietic cells. </li> <li>Participate in team drug discovery biology efforts to perform target validation, discovery toolkit development and hit-to-lead selection and progression. Opportunity to champion projects as they progress to hit-to-lead and beyond.</li> <li>In collaboration with the team:<ul> <li>utilize literature and internal bioinformatics data to design and test hypotheses for drug targets</li> <li>identify pharmacodynamic assays for utility in pre-clinical and clinical setting; define PK/PD/efficacy relationships and test patient selection strategies.</li> </ul> </li> <li>Interpret, present and report research findings at internal meetings. </li> </ul> <p></p> <p><br></p><p><strong>Requirements</strong></p><p>MS or PhD equivalent in Immunology, Cell Biology or Cancer Biology with 3+ years hands-on experience in drug discovery research in a pharmaceutical setting</p><p>Extensive experience with protein detection/quantitation methodology (e.g., Western blotting, JES) and antibody reagent validation required</p><p>Multiparameter flow cytometry and immune cell assay experience required (e.g. T cell proliferation assays); ex vivo cell isolation and analysis experience preferred</p><p>Prior experience preparing drug dilutions and performing drug sensitivity experiments required</p><p>Previous work with culture and maintenance of mammalian cell lines, including transfection and transduction techniques, as well as stable cell line generation; prior experience culturing/manipulating primary immune cells preferred</p><p>Ability to maintain timely and detailed notes using electronic lab notebook as well as demonstrated experience using Microsoft Excel and GraphPad Prism</p><p>Excellent interpersonal skills and willingness to learn a variety of new techniques and biological approaches; ability to work in a matrixed team and adapt to rapidly changing priorities and deadlines</p> Monte Rosa Therapeutics Boston MA
