SUMMARY

Generation Bio is seeking a self-starting, energetic, and versatile (Sr.) Associate Scientist to join our Preclinical Research Team, focusing on the liver platforms. This lab-based position will include both in-vitro and ex-vivo assay work, both routine and development. The successful candidate will have a background with plate-based assays, assay development, and mammalian cell culture. This role will be responsible for implementing cell-based and ex-vivo assays to characterize lead gene therapy candidates for a range of implications.

RESPONSIBILITIES:

  • Care and maintenance of mammalian cell lines for use in in vitro studies.
  • Optimize and execute cell- and plate-based assays to assess expression(i.e., ELISA, MSD, RT-qPCR, Western Blot) & functional activity (e.g. target engagement, enzymatic activity) from tissue samples, plasma/serum, and cell culture.
  • Design and troubleshoot experiments, develop protocols, analyze data and interpret results.
  • Summarize studies in oral and written reports, and present new data across teams.

QUALIFICATIONS:

  • B.S. or M.S. with 1-3 years of relevant laboratory experience outside of coursework. (required)
  • Proficiency in mammalian cell culture, and cell manipulation techniques (e.g. transfection). (required)
  • Excellent laboratory techniques and the ability to work independently to analyze, interpret and present data.
  • Proficient in protein detection methods including ELISA, MSD, and Western Blot analysis from both in vivo (tissue/plasma/serum) and in vitro (cells) samples.
  • Experience with cell sorting/FACS/flow cytometry and/or cell imaging methods (e.g. IHC, ICC/IF) a plus.
  • Experience with gene expression assays, such as RT-qPCR is a plus.
  • Ability to multi-task to support multiple projects.
  • Ability to work independently and analyze and interpret data.
  • Ability to work in a highly collaborative, cross-functional environment.
  • Excellent verbal and written communication skills.
  • Prior experience in gene therapy and/or lead optimization a plus.

For this position, the successful candidate will support our antibody gene transfer platform and preclinical development of our growing pipeline of development candidates. The ideal candidate will have a strong background in research and development of antibody biologics with experience in their design, molecular cloning, expression, and pharmacology. This role will be primarily responsible for in vivo characterization of antibody gene therapies, including designing and executing animal studies to analyze pharmacokinetic properties as well as functional activity of expressed antibodies. This work will also include cross-functional research to optimize our platform for antibody expression and contribute to the identification of new targets and design of therapeutic candidates. RESPONSIBILITIES: * Develop tools and design in vivo experiments to screen lead constructs and characterize antibody PK/PD * Design methods to structurally characterize lead molecules produced in vivo * Design, optimize, and execute assays to assess antibody expression (e.g. RT-qPCR, ELISA, Western Blot) & biological effects (e.g. target engagement, activity, effector function) from tissue samples/plasma/serum * Develop, characterize and evaluate animal disease models * Manage external CRO activities, including project design, timelines and quality of deliverables * Design experiments, analyze data & interpret results * Summarize studies in written reports and oral presentations * Review literature to identify relevant disease targets and therapeutic antibodies REQUIREMENTS: * PhD in Biochemistry, Pharmacology, Molecular and/or Cell Biology, Virology, or related discipline * Minimum of 5 years of industry experience preferred * B.S. or M.S. with 10+ years of relevant industry experience * Excellent lab technique and the ability to work independently to analyze, interpret, and present data * Experience in infectious diseases and execution of viral challenge studies preferred * Proficient in protein detection methods including ELISA and Western Blot analysis from in vivo (tissue/plasma/serum) and in vitro (cells) samples * Experience with DNA/RNA purification and quantification, biochemical assays, enzymatic assays, and/or cell imaging methods (e.g. IHC, FACS) * Ability to multi-task to support multiple projects across platform and disease biology pharmacology * Ability to work in a highly collaborative, team-based environment * Excellent written and verbal communication skills * Prior experience with gene therapy a plus POSITION: Full-Time, Exempt EEOC Statement: Generation Bio believes in a diverse environment and is committed to equal employment opportunity for all its employees and qualified applicants.
