Responsibilities:

  • Responsible for the development, optimization and scale-up of downstream manufacturing processes for large scale plasmid and protein production.

  • Designs and conducts experiments with lab notebook/documentation that follow protocols and accurately records procedure and results.

  • Optimizes protein and plasmid recovery from microbial cells and culture media and protein refolding; screens chromatography resins; optimizes overall purification process, chromatography, ultrafiltration (UF) and diafiltration (DF) conditions; optimizes overall product quality and yield.

  • Completes analytical testing for process science experiments and manufacturing investigations.

  • Prepares components, media, buffers, and other solutions as needed.

  • Performs data analysis and prepares results summaries.

  • Prepares reports and project presentations.

  • Regular and reliable attendance on a full time basis [or in accordance with posted schedule].

  • Responsible for exhibiting professional behavior with both internal and external business associates that reflects positively on the company and is consistent with the company's policies and practices.

Requirements:

  • Bachelor's degree in a Life Sciences field or equivalent required.

  • Minimum of twelve (12) years of experience in protein and plasmid purification.

  • Extensive experience and specialist understanding of principles of anion/cation exchange, hydrophobic interaction, multimodal and hydroxyapatite chromatography.

  • Specialist understanding of principles and extensive experience with tangential flow filtration (TFF), ultrafiltration (UF) and diafiltration (DF).

  • Extensive analytical assay experience (SDS PAGE and agarose gel electrophoresis, protein and DNA concentration determinations, immunoassays)

  • Adept at experimental design and analysis.

  • Protein and/or plasmid manufacturing experience.

(*in collaboration with AbbVie) About the Role: Among other responsibilities, the Associate Scientist will support the design and execution of solid-state material characterization and optimization studies to support product development from early discovery through commercialization phases. In addition, s/he will assure full characterization of incoming API, drug products, and raw materials, provide hands-on analytical support, and packaging and testing of drug products. The Associate Scientist will also assist in screening of polymorph and salt forms to support selection of a developable candidates along with other preformulation characterization studies, maintain laboratory equipment and perform calibrations/verifications as needed. _ Your Contributions (include, but are not limited to): * Characterize physiochemical properties of API and Drug Product materials via DSC, TGA, PXRD, GVS, PLM, and SEM * Measure biopharmaceutical properties, pkA and logP, of materials via UV/Metric and pH/Metric titration techniques * Execute thermodynamic and kinetic solubility studies to aid in method development processes * Perform salt and polymorph screens to enable form selection of developable drug candidates * Assist in studies to understand and optimize crystallizations of a target polymorph * Evaluate powder properties and physical characteristics of API and DP to de-risk later stage DP manufacturing operations. * Operate and maintain laboratory instrumentation and equipment, and facilitate service with technicians as needed * Collaborate and communicate with other groups in-house to ensure conclusions from data generated are applied correctly and used widely and effectively * Assemble and maintain documentation of data to allow efficient retrieval of results * Performs other duties as assigned. Requirements: * BS/BA degree in Chemistry, Chemical Engineering, Materials Science, or related field and some experience in a chemistry, analytical, or characterization laboratory setting required; Experience with characterization equipment such as PXRD, DSC, TGA, and SEM a plus, OR * MS/MA degree in Chemistry, Chemical Engineering, Materials Science, or related field preferred and some lab or industry experience (preferred) * Some knowledge of a cGMP-regulated environment, including familiarity of relevant US/EU regulatory and quality requirements, standards, and practices. * Familiarity with biotech/pharma manufacturing operations, product and process development, finance, and supply chain operations * Possess familiarity of physical & analytical techniques as they pertain to biopharmaceutical properties. * Ability to learn quickly and think independently. * Familiarity with the development and execution of chemical and instrumental methods (e.g. HPLC, Solubility Analysis, DSC, TGA, PXRD, Particle-Size Analysis, GVS, PLM, T3, and SEM) of analyses for development compounds, drug substances, raw materials, intermediates, drug products, and packaging materials. * Able to execute experiments efficiently and suggest small process improvements * Able to recognize patterns or inconsistencies in data results * Able to deduce meaningful results while conducting routine analyses or troubleshoot minor issues.
