I: Job Summary

The Assistant Scientist in the Molecular Biology and Protein Engineering group will play a critical role in the development and optimization of protein reagents and drug candidates based on the ADAPTIR platform, including monospecific and bispecific antibodies. Primary activities include plasmid vector design and cloning, DNA sequencing analysis, and phage display, with opportunities to grow into more advanced molecular engineering projects.

II: Responsibilites

  • Utilize standard molecular biology techniques, including bacterial cloning, DNA production and purification, site directed mutagenesis, Gibson Assembly, primer design, PCR and agarose gel electrophoresis to produce expression vectors encoding protein reagents and protein therapeutics

  • Participate in phage display panning to isolate new or optimized binding domains to targets of interest

  • Recombinant protein expression and characterization in in vitro assays

  • Cell culture and aseptic technique

  • Database entry and maintenance

  • Documentation of experiments and results in laboratory notebooks

  • Provide lab support while working in a collaborative team environment

III: Education, Experience & Skills

  • 4 year college degree in Biology, Biochemistry or a related life sciences field

  • 1 - 3 years of relevant academic or industrial experience in molecular biology or protein engineering is required.

  • Demonstrated competency in standard molecular biology techniques.

  • Experience with DNA software tools for plasmid and PCR sequence analysis (e.g. Vector NTI)

  • Outstanding organizational and record keeping skills, with a demonstrated ability to multi-task in a fast-paced environment.

  • Ability to meticulously maintain a lab notebook.

  • Excellent communication skills, independently motivated, detail oriented, and demonstrate good problem solving ability.

  • Familiarity with phage display a plus

IV: Physical/Mental Requirements

Type/keyboard, visual acuity, good eye/hand coordination, stand, walk, sit, twist/turn, climb, reach outward, reach above shoulder, hand dexterity, bend, squat/kneel, sense of feel, sense of hearing, sense of smell, lift/carry 25 lbs, and pull 25 lbs. Use phone, fax, copier, and computer. Use hand tools and operate fork lift. Organize/coordinate, analyze/interpret, calibrate precise measurements, problem solve, make decisions, supervise, plan, communicate, prepare written communications, and prepare transaction documents. Drive automatic/standard vehicle. Work outdoors, around noise above conversation level, in hot/cold temperatures, in high humidity, where ventilation is needed, in multiple locations, with restricted access to laboratory, in contact with Select Agents, and with exposure to animals, animal areas, or equipment used in animal areas.

They Independently develop and follow protocols and function with minimal supervision. She or he works on complex issues where analysis of situations or data requires in-depth evaluation of variable factors. They exercise judgment in selecting methods, techniques and evaluation criteria for obtaining results. They network with key contacts outside own area of expertise to accomplish tasks that impact outside R&D departments. They determine methods and procedures on medium- to large-scale experimental assignments and may coordinate activities of other personnel (Team Lead). Some barriers to entry exist at this level (e.g., dept / peer review). Level at which career may plateau. Typically requires a university degree and a minimum of 8 years of related experience. At this level the ability and experience should be considered as post-graduate coursework if comparable. Postgraduate coursework may be desirable on paper. Primary Responsibilities * Develop new PCR-based assays for immune system profiling by next generation sequencing in collaboration with a team of researchers and on the nexus to process engineering, computational biology, and production service laboratory. * Create, develop and introduce new methods and technologies to advance research projects. * Work with validation scientists, process engineers, computational scientist, and Production team to build sensitive and robust PCR-based assays for both manual and automated systems. * Provide expertise in selecting, designing, optimizing, qualifying, validating and implementing molecular assays and laboratory platforms for research and clinical applications from research to production setting. * Emphasize purpose-driven decision making with positive long-term impact through technical and practical implementation. * Design experimental procedures and search literature for targeted information. * Record experimental details and maintain laboratory notebook. Other characteristics * Interact with manager and project team to review data and plan next steps. * Cultivate positive and productive working relationships with colleagues. * Prepare and present work at group meetings. * Flexible to changing priorities. * Respectful of laboratory safety policies and practices. * Review and analyze data, interpret results, report findings to supervisor; form hypothesis, define next course of actions and design well-controlled experiments to address the question of interest. Position Requirements Education * Bachelor degree or masters degree in biology, molecular biology, immunology, or similar discipline or PhD degree within a scientific discipline or equivalent in engineering. Years of experience * Minimum of 7+ years of laboratory experience. Advanced degree may be substituted for experience as appropriate. Specialized Skills * Knowledge of standard lab practices, particularly that of a BSL-2. * Expert knowledge of PCR platforms. * Extended knowledge of liquid handling platforms with extensive experience designing and implementing molecular assay Knowledge in assay validation, design purpose driven experiment considering parameters including but not limited to disease type, patient population, tissue types, sample matrix, sampling method and storage condition, extraction method, sensitivity and dynamic range of the assays and analytical platforms, purpose of analyte measurement (qualitative, or quantitative) etc. * Familiarity with command line tools and simple programming tasks in python, perl, or R a plus but not required, but willingness to learn necessary. * LIMS experience a plus but not required. * Windows OS, Mac OS, and MS Office literacy required. Physical Requirements * Finger/hand dexterity, endurance, and mental focus are required for successfully carrying out wet lab work. * Ability to walk through labs, sit or stand for long periods while performing lab work. * Occasional lifting of heavy equipment. * Requires work in BSL-2 labs, following all guidelines and protective measures.
