I: Job Summary

The Assistant Scientist in the Molecular Biology and Protein Engineering group will play a critical role in the development and optimization of protein reagents and drug candidates based on the ADAPTIR platform, including monospecific and bispecific antibodies. Primary activities include plasmid vector design and cloning, DNA sequencing analysis, and phage display, with opportunities to grow into more advanced molecular engineering projects.

II: Responsibilites

  • Utilize standard molecular biology techniques, including bacterial cloning, DNA production and purification, site directed mutagenesis, Gibson Assembly, primer design, PCR and agarose gel electrophoresis to produce expression vectors encoding protein reagents and protein therapeutics

  • Participate in phage display panning to isolate new or optimized binding domains to targets of interest

  • Recombinant protein expression and characterization in in vitro assays

  • Cell culture and aseptic technique

  • Database entry and maintenance

  • Documentation of experiments and results in laboratory notebooks

  • Provide lab support while working in a collaborative team environment

III: Education, Experience & Skills

  • 4 year college degree in Biology, Biochemistry or a related life sciences field

  • 1 - 3 years of relevant academic or industrial experience in molecular biology or protein engineering is required.

  • Demonstrated competency in standard molecular biology techniques.

  • Experience with DNA software tools for plasmid and PCR sequence analysis (e.g. Vector NTI)

  • Outstanding organizational and record keeping skills, with a demonstrated ability to multi-task in a fast-paced environment.

  • Ability to meticulously maintain a lab notebook.

  • Excellent communication skills, independently motivated, detail oriented, and demonstrate good problem solving ability.

  • Familiarity with phage display a plus

The individual is expected to work with a dynamic team of individuals towards a common goal. The Scientist will conduct tasks and assignments as directed and contribute to team objectives and outcomes as guided. The Allen Institute creates broad and useful data and analytical tools for the scientific community. The Scientist Computational Biology/Immunology brings scientific and technical expertise to a new initiative at the Allen Institute that will focus on understanding human immunology. We will be applying high resolution molecular biology technologies and state of the art immunological analyses at the single cell level to achieve our goals in longitudinal studies of normal humans and in human diseases over multiyear periods. The Scientist for Computational Biology/Immunologyy will be a critical part of the team. This individual provides hands-on contributions to the analyses of complex data sets derived from a suite of high resolution technologies including single cell transcriptomics, flow cytometry, CYTOF on multiple cellular lineages in the immune system and hypothesis based experimental immunology efforts. He or she will integrate high-throughput experimental assays with computational pipelines, develop novel data visualization and analysis techniques/tools and contribute to future research hypotheses and directions. As a member of the computation biology group, the Scientist will work closely with other team members including an experimental immunology group, a high-resolution biology group, and an integrated information technology team. Extensive collaborations with external academic partners are anticipated to carry out project goals and objectives. The ideal candidate would have documented experience applying quantitative, computational and single cell molecular biology techniques in prior investigations of the human immune system including longitudinal data analysis. Job Responsibilities: * Contribute as a scientist in a dedicated computational biology group studying human immunology * Carry out a combination of benchwork and computational work supporting the research pipeline for Immunology to include lineage/trajectory modeling, transcriptomic based clustering, and data integration with both preclinical and clinical data bases aimed at understanding immune health and disease mechanisms * Coordinate and manage tasks as assigned and identify new areas where action is required. Bring new concepts in bioinformatics and computational tools to the organization. Provide solutions by new tool development. Be an active driver of new hypotheses in concert with other groups in Immunology * Assist, train, and mentor other members of the immunology team and external partners, as needed * Prepare written and oral reports and/or publications with accuracy and reliability. Assist in the development and documentation of standardized operating procedures that will be shared with collaborators and the broader immunology community. * Serve as a computational biology expert and consultant to both internal and external collaborators * Demonstrate integrity, credibility and a positive attitude in daily performance to motivate others to do the same. * Other duties as required. Knowledge/Skills/Abilities * Excellent written and verbal communication skills. * Demonstrated ability to document work accurately and precisely. * Strong organizational skills and careful attention to detail. * Able to work independently and in a collaborative high paced environment * Able to meet departmental goals and milestones across multiple projects. Required Job Qualifications * Ph.D. in mathematics, computational biology, biostatistics or related disciplines * 2 or more years of relevant post Ph.D. experience. * Experience in immunology related programs or mammalian cell signaling/biochemical pathways * Strong familiarity with contemporary bioinformatic tools and hands on tool development expertise * Experience with single cell isolation, lineage tracking techniques and population modeling. * Experience in the analysis of both single cell or populations a plus; Mass spectrometry(CYTOF), high-throughput cell cytometry, cell sorting for single cell RNA sequencing applications. * Experience working in a cross disciplinary, team based environment in academia or industry; and working with outside collaborators to achieve defined objectives. * Authorship demonstrating capabilities in top tiered peer reviewed journals is a plus. Preferred Job Qualifications * Ability to develop, test, implement, and share new experimental and computational tools quickly, in an iterative manner, after feedback from experimental, data production, and analysis teams. * 2+ years of experience studying human immunology * Exceptional oral and written communication skills. * Ability to work independently as well as part of a team to meet aggressive timelines in a collaborative environment.
